Course Description: This course is designed to give practical experience for graduate students in learning, understanding and applying molecular biology techniques for their own research projects. The students will extract the DNA from their own invertebrate or pathogen of interest. That material will be used for further experiments involving restriction endonuclease (REN) digestions, DNA cloning, DNA Southern blotting, PCR amplification, DNA sequencing and DNA data analysis using GCG computer programs, phylogeny using PAUP and other programs. Students will also learn cell culture handling procedures. Lectures will provide the fundamental background necessary for understanding molecular biology pertinent to the methodology that will be used. Since new molecular methodologies are constantly introduced and modified, the course techniques will be updated as needed.
Prerequisites: Any basic course in genetics, biochemistry or molecular biology.
Textbook: None. Protocols and handouts will be given.
Assignments and Methods by which the Student will be Evaluated and Grades Determined: The course grade will be determined by how well the student maintains the laboratory notebook (30%), preparation of protocols (10%), experimental results (20%), participation and discussion (15%) and the presentation of an experimental project involving molecular techniques (25%).
Critical Dates for Exams or Other Work: There are no written exams. The student project presentations will be scheduled during the semster.
Policy Related to Class Attendance: Students are expected to attend all class sessions. Experimental procedures each day build upon the next for manipulation of DNA.
Class Demeanor Expected by Instructor: Please be considerate of your classmates by not chatting during lecture. The banging of doors is very distracting to both students and professor, therefore please arrive on time and do not leave early. Turn off cell phones and beepers before coming into classroom. UF rules prohibit your having food or drinks in classrooms. Use of tobacco products (in any form) in the classroom is prohibited.
